High-Productive Cell Separation by Dielectrophoresis in Free Flow

A construction of a stationary device and method for separation of microbiological samples on the basis of dielectrophoresis in the free flow have been suggested. The principle underlying the method is the formation of dielectrophoretic barriers in the volume of separation chamber which enhances the productivity of the process and makes it simpler as compared to the existing conventional systems of planar dielectrophoresis. The computer modeling of distribution of electric fields intensities was performed, and the areas of dielectric barrier formation were determined. The effect of forces on a particle (yeast cell 7 μ in diameter) in the separation chamber was assessed using the data of the computer modeling. The high efficiency of Saccharomyces cerevisiae yeast cells separation by the device (> 99%) and its high productivity (2.5 cell/s) were theoretically predicted and experimentally confirmed.
